Description and Applications

SP6 RNA Polymerase is a DNA-dependent RNA polymerase which has stringent specificity for the SP6 phage promoters. This property enables SP6 RNA Polymerase to efficiently synthesize in vitro transcripts from almost any DNA that is downstream from a SP6 promoter. These transcripts have proven to be useful as hybridization probes and as substrates in RNA processing systems.

One unit is the amount of enzyme required to incorporate 1 nmole of labeled UTP into acid-insoluble material in 60 minutes at 37°C

Assay Conditions

40 mM Tris-HCl (pH 7.9)

4.0 mM spermidine-(HCl)3
1.0 µg SP6 DNA
0.04 mM of each ATP, CTP, GTP and [α-32P] UTP
Incubation is for 10 minutes at 37°C in a total reaction volume of 50 µl
