CMX Buffer 4
Unit Definition
One unit is the amount of enzyme required to digest 1 µg of lambda DNA with ≥95% efficiency in 1 hour in a total reaction volume of 50 μl
Incubation Temperature
37°C
Heat Inactivation
10 minutes at 65°C
Reaction Buffer
20 mM Tris-Acetate (pH 7.5 at 37ºC)
50 mM Potassium Acetate
10 mM Magnesium Acetate
1 mM dithiothreitol
100 µg/ml bovine serum albumin
Storage Buffer
10 mM Tris‑HCl (pH 7.5 at 4ºC)
0.1 mM EDTA
1 mM dithiothreitol
50 mM KCl
200 µg/ml bovine serum albumin
50% (v/v) glycerol
Assay Conditions
20 mM Tris‑Acetate (pH 7.5 at 37ºC)
50 mM Potassium Acetate
10 mM Magnesium Acetate
1.0 mM dithiothreitol
100 µg/ml bovine serum albumin
1 µg lambda DNA
Incubation is at 37°C for 1 hour in a total reaction volume of 50 µl
Notes
- Lambda contains 3 slow-cleavage sites that do not completely digest after 1 hour incubation
- Complete digestion of these sites is achieved by 4 Units of Rsr II with 1 μg of lambda DNA incubated at 37°C for 16 hours
Store at -20°C
Shipped on dry ice
