20 mM Tris-Acetate (pH 7.5 at 37ºC)
50 mM Potassium Acetate
10 mM Magnesium Acetate
1 mM dithiothreitol
100 µg/ml bovine serum albumin

Storage Buffer

10 mM Tris‑HCl (pH 7.5 at 4ºC) 
0.1 mM EDTA
1 mM dithiothreitol
50 mM KCl
200 µg/ml bovine serum albumin
50% (v/v) glycerol

Assay Conditions

20 mM Tris‑Acetate (pH 7.5 at 37ºC)
50 mM Potassium Acetate
10 mM Magnesium Acetate
1.0 mM dithiothreitol
100 µg/ml bovine serum albumin
1 µg lambda DNA
Incubation is at 37°C for 1 hour in a total reaction volume of 50 µl

Notes

1. Lambda contains 3 slow-cleavage sites that do not completely digest after 1 hour incubation
2. Complete digestion of these sites is achieved by 4 Units of Rsr II with 1 μg of lambda DNA incubated at 37°C for 16 hours
