Agarase

Description

• Potent enzyme that digests the polysaccharide backbone of agarose yielding ethanol soluble oligosaccharides.
• Resulting carbohydrate molecules no longer gel or interfere with subsequent DNA manipulations.
• Ideal for quantitative recovery of high molecular weight DNA from low-melting agarose gels.
• Simple, quantitative recovery of intact nucleic acids from agarose gels.
• May be used to purify large (>50 Kb) and small (<50Kb) DNA fragments from agarose gels.
• DNase- and RNase-free.
• Can be heat inactivated (2 minutes at 95°C or 15 minutes at 65°C).

Unit Definition

One unit is the amount of enzyme required to complely degrade 200 ul of molten 1% agarose in reaction buffer in 1 hour at  41°C.

40 mM Tris-HCl (pH 7.5)
0.5 mM EDTA
100 mM NaCl
1 mM dithiothreitol 0.1% Triton X-100™
50% glycerol 

Reaction Buffer

40 mM Bis-Tris (pH 6.0)
40 mM NaCl
0.5 mM EDTA

Assay Conditions

50 mM Tris- HCl (pH 7.6)
10 mM MgCl₂ 
1.0 mM dithiothreitol
0.15 mM duplex [³³P] lambda DNA
Incubation at 37ºC for 30 minutes in a reaction volume of 20 μl.
