Unit Definition

One unit is the amount of enzyme required to incorporate 10 nmoles of total nucleotide into acid-insoluble material in 30 minutes at 70°C

Storage Buffer

50 mM Tris-HCl (pH 7.5)
5.0 mM dithiothreitol
0.1 mM EDTA
50% (v/v) glycerol

Assay Conditions

25 mM Tris-HCl (pH 9.5)
10 mM MgCl₂
1.0 mg/ml bovine serum albumin
0.2 mM of each dCTP, dGTP, dTTP and [α-³²P] dATP
15 µg activated DNA
Incubation for 30 minutes at 37°C in a total reaction volume of 100 µl
